E-TPGS® Natural Vitamin E TPGS:

D-α-Tocopheryl polyethylene glycol succinate (Vitamin E TPGS, or simply TPGS) is a water-soluble derivative of natural Vitamin E, formed by esterifying Vitamin E succinate with polyethylene glycol (PEG). This unique amphiphilic structure, with a lipophilic alkyl tail and hydrophilic polar head, has a hydrophile/lipophile balance (HLB) of 13.2 and a low critical micelle concentration (CMC) of 0.02% w/w. These properties enable stable micelle formation, enhancing supplement solubility, bioavailability, and stability in various formulations.

E-TPGS® combines the benefits of PEG and Vitamin E, making it a valuable excipient in nanocarrier-based drug delivery. It extends a drug’s half-life in plasma, improves cellular uptake, and acts as a P-glycoprotein (P-gp) inhibitor to help overcome multidrug resistance (MDR) in cancer therapy. Its versatility extends to pharmaceutical, nutritional, and cosmetic applications, with ongoing research exploring its potential in targeted nanomedicines.
